The Basic Science Program pursues independent, multidisciplinary research programs. Research efforts and support are an integral part of the Center for Cancer Research (CCR) at the Frederick National Laboratory for Cancer Research. This position is in the laboratory of Dr. Ruth Nussinov, a preeminent computational structural biologist. A postdoctoral fellowship in computational structural biology with Dr. Ruth Nussinov is available for a project at the Frederick National Laboratory for Cancer Research.

The project aims to decipher functional mechanisms of proteins to discover the structural basis of disease. It involves simulations of oncogenic proteins aiming to figure out their activation mechanisms, the actions of their driver mutations, signaling, allosteric drug discovery, and the emerging connection to neurodevelopmental disorders. We are studying a population of multipotent axial progenitors, called neuromesodermal progenitors (NMPs), which arise during gastrulation and generate the spinal cord and somites of the trunk and tail. The Wnt/beta-catenin signaling pathway activates gene regulatory networks (GRNs) to regulate NMP development.

Summary of Postdoctoral Fellowship:

A postdoctoral fellowship position is available in the laboratory of Natalie Porat-Shliom. Ph.D., within the Center for Cancer Research (CCR), NCI, NIH, in Bethesda, Maryland. Candidates are sought to join a basic and translational research program focused on hepatocytes cell biology and non-alcoholic fatty liver disease. Our lab is interested in fundamental questions related to mitochondrial biology in the liver.

How are mitochondrial structure and function regulated? What are the environmental cues/stressors that induce these mitochondrial alterations? And, in turn, how are those disrupted in human disease? For this, we use intravital microscopy, the study of cellular processes in live, anesthetized rodents (Cunningham and Porat-Shliom 2021, Stefkovich et al 2021, Porat-Shliom et al. 2019). A benefit of this position is the opportunity to learn state-of-the-art light microscopy imaging techniques.

A post-doctoral position is available in the laboratory of Avinash Bhandoola at the National Cancer Institute. The laboratory studies the development of T cells, innate lymphocytes, and thymic epithelial cells. New cellular intermediates in the development of innate lymphocytes were recently described by Dr. Bhandoola’s lab, and we wish to evaluate the role of specific transcription factors in this process. Other work in the laboratory studies regulation of thymus growth, and is focused on identifying new types of thymic epithelial cells and transcription factors important for their function.

MicroRNAs (miRNAs) play critical roles in both mammalian development and human diseases such as cancer. The goal of our program is to study how miRNAs themselves are regulated, and to apply gained insights into developing effective treatments for cancer. Using genetic studies in living cells and animals, together with biochemical and next-generation sequencing (NGS) approaches, our laboratory focuses on elucidating mechanisms of miRNA biogenesis and the roles that miRNAs play in tumorigenesis.

Postdoctoral positions are immediately available in the laboratory of Dr. Shiv Grewal, Laboratory of Biochemistry and Molecular Biology, CCR, NCI, NIH. The Grewal lab aims to uncover fundamental principles governing the assembly and epigenetic inheritance of heterochromatin, a repressive form of chromatin that controls gene expression and protects genome stability.

The major focus of the lab is on understanding how highly conserved RNA-based mechanisms and chromatin modifying activities assemble heterochromatin at specific sites across the genome, and how repressive chromatin structures are epigenetically inherited. Current projects seek to define the roles of novel RNA processing factors in the formation of large heterochromatin domains and uncover the mechanisms by which modified histones provide an epigenetic template to propagate repressive chromatin, which is critical for stable maintenance of gene expression patterns during development.

In other exciting areas of interest, the lab is exploring how heterochromatin assembly impacts 3D genome organization and investigating adaptive genome control, in which heterochromatic structures are dynamically modulated in response to environmental or developmental cues to allow appropriate gene expression profiles.

The Protein and Chromatin Folding Section, Laboratory of Biochemistry and Molecular Biology, CCR, NCI, NIH is recruiting a postdoctoral fellow. Candidates with an interest in chromatin structure and its relationship to cancer are encouraged to apply. The DNA molecules in eukaryotic cells are associated with histone proteins to form chromatin that can fold to various structures for the purpose of biological functions.

The folding of chromatin is regulated by many other non-histone protein factors. We study chromatin folding through determination of the structures and dynamics of chromatin units and their complexes with regulatory proteins using single particle cryo-EM, X-ray crystallography, solution NMR, and other biophysical/biochemical methods.

Our most recent studies include the high-resolution structural determination of chromatosomes with different linker histone isoforms, centromeric nucleosomes bound to kinetochore proteins, and nucleosomes with native DNA sequences and how they are recognized by pioneer transcription factors.
