At Stanford, innovations in neural interface technology have allowed us to discover how abnormal electrical brain activity contributes to disorders in movement. In the Human Motor Control and Neuromodulation Lab, the first decoding of electrical activity in deep brain structures during abnormal movement in Parkinson’s disease patients was performed using novel and investigative sensing neurostimulators.
Our team has deconstructed brain activity to discover both the neural code responsible and kinematic quantification for various motor symptoms in Parkinson’s disease. We are interested in understanding how altered cellular metabolism drives tumorigenesis and identifying metabolic liabilities amenable to therapeutic targeting. Recently, the oncometabolite hypothesis of cellular transformation has emerged as a central mechanism of oncogenesis. According to this concept, excess accumulation of intermediary metabolites such as succinate, 2-hydroxygluterate, and fumarate that result from altered succinate dehydrogenase (SDH), isocitrate dehydrogenase (IDH) or fumarate hydratase (FH) function, respectively, drive pathogenic epigenetic cellular reprogramming and tumor formation.
However, model systems to interrogate the oncometabolite hypothesis and identify therapeutic targets are lacking. To address this critical need, the successful applicant will develop new (human and mouse tumor-based) and leverage established (PMID: 35235785) models of succinate dehydrogenase (SDH)-deficient tumorigenesis. These models will be used for mechanistic investigation and to establish high throughput chemical and genetic discovery platforms. This work will have substantial impact on our fundamental understanding of tumor formation and uncover new treatment avenues.

Neuronal Signal Transduction Research: There is an open postdoctoral position in the laboratory of Michael S. Kapiloff, MD, PhD, Reinhard Family Professor in the Departments of Ophthalmology and Medicine. We are seeking to recruit a highly motivated cellular and molecular biologist with interest in neuronal signal transduction. Current research in the Kapiloff lab addresses how the formation of multimolecular protein complexes confers specificity in intracellular signaling and how these complexes are important for the survival of central nervous system neurons in diseases such as glaucoma and stroke.
A major focus of the lab has been the mAKAP scaffold protein that coordinates crosstalk between cAMP, calcium and MAP-kinase signaling pathways and that is important for neurite extension and neuroprotection in the eye. Basic research concepts studied in the laboratory are being applied to the development of novel gene therapy vectors for the treatment of retinal diseases (glaucoma). This position will involve extensive collaboration with the laboratory of Dr. Jeffrey L. Goldberg, Chairman of Ophthalmology at Stanford. In particular, this position involves the study of signaling pathways in living cells and tissues using fluorescent biosensors.

The Mignot lab studies autoimmune neurological diseases with a focus on immunogenetics and functional autoimmunity.
