Research in the Voss laboratory is centered on the development of engineered B cell therapies to achieve an HIV functional cure. A position is immediately available for a highly motivated individual who is interested in improving B cell genome-editing approaches and characterizing the function of engineered cells in vivo in the mouse and non-human primate (NHP) animal models. The successful candidate will join a small team that will include doctoral students and research assistants who will work together to accomplish experimental goals. The Ghadiri Laboratory at the Scripps Research, Department of Chemistry, and the Skaggs Institute for Chemical Biology is currently seeking talented Postdoctoral Associate candidates with a strong background in biology, molecular biology, chemical biology, and/or biochemistry. The lab is developing new, general approaches to control the site and timing of enzyme activity. Enzymes have great potential as therapeutic and diagnostic agents due to their high specificity and catalytic activity, but the ability to better control the timing and location of enzyme function would enable the use of a wider variety of enzymes for therapeutic purposes. Our approach for enzyme programming relies on attaching an enzyme to its small-molecule inhibitor through an allosteric DNA tether. Upon interaction of the DNA tether with its complement strand, the ensuing conformational change activates the enzyme by dislodging the inhibitor from its active site. We have established the utility of DNA-programmed intrasterically-regulated enzymes in developing constructs for fast, sensitive, and PCR-independent isothermal detection of label-free nucleic acid sequences; carrying out complex logic operations; programming the activity of streptokinase, a clinically-used fibrinogen activator; and specifically killing targeted cancer cells. We are seeking qualified candidates who can join our multidisciplinary team of scientists to lead this broad research effort.

Hepatocellular carcinoma (HCC) is a major cause of cancer death worldwide, accounting for over half a million deaths per year. Chronic infections with hepatitis B (HBV) and C (HCV) viruses have been recognized as main human liver carcinogens. Multiple non-viral factors have also been implicated in the development of HCC. The development of non-alcoholic steatohepatitis (NASH) and liver fibrosis represent high risk factors for HCC. Remarkably, we recently found that cyclophilin inhibitors suppress the development of viral-induced and non-viral-induced liver fibrosis and HCC in mouse models. However, their mechanisms of action are totally unknown. A postdoctoral position is immediately available to study the liver damage protective effects and mechanisms of action of cyclophilin inhibitors, both in vivo and in vitro, and to determine the potential of cyclophilin inhibitors as anti-liver damage drugs.

SARS-CoV-2 is currently a major cause of death worldwide. Although vaccines are emerging, new therapies are urgently required. We recently developed a novel approach that permits the killing of SARS-CoV-2-infected cells while preserving non-infected cells. This new approach exploits the viral replication machinery (specifically the SARS-CoV-2 polymerase) to induce the expression of killer genes (diphtheria toxin fragment A) which triggers a rapid specific death of viral-infected cells, but not non-infected cells. We are interested in studying RNAs which are expressed only upon gain of function in a driver oncoprotein. These lncRNAs are candidate markers of oncogenic transformation and in some cases may contribute to the oncogenic phenotype. An important goal of our work is the identification of lncRNAs that possess growth-promoting effects on cancer cells. As a tool, we use targeted Cas9 libraries. Once identified, the lncRNAs are characterized both functionally and structurally.
