We have two, fully funded positions, with either a focus on experimental immunology or quantitative genetics and data analyses. Dr. Lingbo Zhang’s Lab at Cold Spring Harbor Laboratory focuses on both basic science and translational medicine of hematologic malignancies. The research of the Lab centers on normal and malignant stem and progenitor cells in the hematopoietic system and decodes the role of metabolites, including micronutrients and neurotransmitters, in the tumor microenvironment and their genetic effectors in regulating hematologic malignancies. There are two fully funded postdoctoral fellowships available to investigate the roles of these critical metabolites and their genetic effectors in hematopoietic malignancies and develop novel therapeutic strategies. The Lab utilizes a combination of CRISPR/Cas functional genomics and metabolomics approaches to systematically uncover the critical metabolites and their genetic effectors, including receptors and metabolic enzymes, to reveal mechanistic insights of and identify key drug targets for hematologic malignancies. The ultimate goal is to understand how environmental signals such as diets and nervous system activities modulate blood cell development and cancers. We are currently seeking innovative, inventive, and self-driven Postdoctoral Associates within the lab of Dr. Leemor Joshua-Tor at Cold Spring Harbor Laboratory. The Joshua-Tor lab studies the structure and mechanism of gene silencing pathways as well as DNA replication. To get a complete understanding of how these processes, the team employs structural, biochemical, biophysical and molecular biology approaches to study how key proteins work at the molecular level. We are interested in how small RNAs identify and silence transposable elements when they become active during mammalian development and disease. We found that tRNA fragments (tRF) derived from the 3’-end of mature tRNAs are highly expressed in cells undergoing epigenetic reprogramming and target LTR-retroelements, or endogenous retroviruses (ERVs), at their conserved tRNA primer binding site (PBS). Due to the perfect sequence complementarity of 3’-tRFs to the PBS, they have tens of thousands of targets in mammalian genomes. 3’-tRFs are processed under yet unknown conditions and potentially protect many eukaryotes. This is an exciting new field with many opportunities! We believe tRFs are an ancient link between RNAi, transposons and genome stability. We are determining the intersection of tRFs with other small RNA silencing pathways and their effect on pluripotency and differentiation. Our main tools are cell culture transposition assays, long & small RNA sequencing, chromatin IPs, NGS analysis of repetitive sequences, CRISPR, etc.
