Summary of Postdoctoral Fellowship:

Our lab investigates how genetic and microenvironmental factors interplay to regulate tumorigenesis. Our research focuses on identification of the cell of origin and elucidating the role of tumor microenvironment in cancer development. We dissect the cellular and molecular mechanisms of carcinogenesis from a developmental perspective using Neurofibromatosis Type 1 (NF1), a common tumor predisposition human genetic disorder, as a model. While working on neurofibromatosis, our research group serendipitously uncovered the identity of follicular epithelial cells that directly give rise to hair and mechanisms that cause hair to turn gray – findings that led to our second focus to identify possible treatments for balding and hair graying associated with aging.

The Zhang laboratory combines a unique mouse forward genetic screening approach with biochemistry and molecular biology to identify new regulators of major metabolic diseases, including obesity, diabetes, and nonalcoholic fatty liver disease (NAFLD). Since the establishment of our laboratory in 2020, our work has led to publications in prestigious journals including PNAS, Nature Communications, and Cell Metabolism. Our laboratory has several exciting projects related to: 1) The dynamic regulation of a newly identified E3 ubiquitin ligase in insulin signaling. 2) The role of several new genes in the regulation of obesity. 3) The mechanism of several new genes in the pathogenesis of NAFLD. A postdoctoral research position is available in the laboratory of Yunsun Nam, in the Department of Biochemistry at University of Texas Southwestern Medical Center in Dallas, to study the mechanisms of noncoding RNAs. Our laboratory works on exciting projects dissecting the molecular mechanisms underlying RNA-mediated gene regulation. To form specific RNA-protein complexes important for normal gene expression, RNA utilizes both chemistry and structure. Thus, we use biochemical and biophysical methods to study how proteins and RNAs recognize each other. More specifically, we investigate core and regulatory mechanisms that govern RNA processing (maturation), modification (eg. methylation) and folding (via helicases) that impact the RNA function in the cell. The noncoding RNAs and RNA/protein complexes we study are relevant for normal development as well as diseases. Our current focus is to unravel how RNA-mediated gene regulation gone awry leads to cancer, and we use our mechanistic understanding to identify therapeutic avenues.
