How insects choose their food sources and how plants can defend themselves are questions of substantial ecological, evolutionary and economical relevance. We seek to provide new insights into these questions by investigating an understudied aspect of plant feeding that all herbivores have in common: the mechanical breakdown of tough plant material. Plant fracture requires the application of a force, the magnitude of which depends on the morphology of the fracturing `tool’, and the material properties of the food source. This force is generated by muscle, in a process that consumes energy. The objective of the project the post holder will carry out is to determine the HLA-associated immunological mechanisms that drive natural protection to type 1 diabetes (T1D) in individuals with the protective DR15-DQ6 haplotype in the presence and absence of known HLA-associated risk alleles (DR4-DQ8 and DR3-DQ2 haplotypes). Single nucleotide polymorphisms in the DR15-DQ6 haplotype are associated with T1D protection. However, 1 in 50 individuals carrying these polymorphisms progress to T1D, representing failure in the genetic protection provided by DR15-DQ6. Understanding the underlying mechanisms by which DR15-DQ6 exerts its protective effects and how this is attenuated in people who carry the variant but still develop T1D would provide invaluable information for the design of therapies that mimic natural protection to T1D. In particular, we will be examining the frequency and functionality of autoantigen-specific CD4+ T cells in people with T1D that harbor risk and/or protective HLA alleles and diabetes-free relatives with similar HLA genotypes.

The research programme in molecular bacteriology, led by Prof. José R Penadés, will analyse the impact of phages in the emergence of antibiotic resistant bacteria. Bacteriophage-mediated gene transfer is a major evolutionary force occurring by two well-described mechanisms: generalized and specialized transduction. We have recently discovered that Staphylococcus and Salmonella phages engage in lateral transduction, the most powerful mode of transduction described to date. In this project, analysing important pathogens, we will investigate whether this ground-breaking strategy of gene transfer is widespread in nature. We anticipate that the results of this study will challenge basic and fundamental concepts of phage biology, which will be revisited here. Moreover, the proposed research has the potential to fundamentally restructure our understanding of the roles of bacteriophages in bacterial evolution, including the development of antimicrobial resistance and virulence in clinical strains.

We are seeking an excellent researcher to work on the development of PCR and sequencing strategies for environmental surveillance samples, with a particular focus on typhoid, paratyphoid and non-typhoidal Salmonella. In preliminary work, we have developed a novel primer scheme to amplify Salmonella Typhi genomic regions that define the genotype and encode antimicrobial resistance (AMR). We are funded by BMGF to optimise this amplicon sequencing approach in silico and in the laboratory, and to extend it to other Salmonella serovars. Summary of Postdoctoral Fellowship:

This research aims to develop novel transition metal coordination compounds as part of new routes to crop yield enhancement. This avoids genetically engineering crops and is based upon the fact that photosynthesis is a surprisingly inefficient process (only ~1-2% efficient). We aim to increase photosynthetic efficiency using wholly novel bio-mimetic methodologies. Some species of plant have developed CO2 concentrating mechanisms that are used to increase cellular CO2 levels.

One of these mechanisms, based upon the catalytic conversion of bicarbonate salts into CO2 by carbonic anhydrases (CAs), has been used as the inspiration for this work. CA has one of the fastest known enzymatic catalytic activities, with a rate of CO2 hydration of ~106 molecules per second per enzyme, and thus has attracted much attention, not only in studies of physiological and biochemical processes, but also as a bio-mimetic template with applications in the pharmaceutical, biotechnological and carbon capture industries.
