Summary of Postdoctoral Fellowships:
This project aims at answering the question: How accurately can one measure low gas pressure in a simple and absolute manner? To attempt to answer it, optomechanical pressure sensors—consisting in highly tensioned strings sculpted on ultrathin dielectric drums—will be realized with the goal of performing absolute pressure measurements with sensitivities beyond the state-of-the-art by monitoring the pressure-induced modifications of the string vibrations (frequencies, damping) with a laser. The project is based on very recent breakthroughs realized by the group regarding the design, fabrication and application of nanostructured silicon nitride thin films. It will involve the use of nanofabrication technologies (Electron beam lithography, atomic force microscopy, etc.) and optical techniques (e.g. interferometry) as well as the simulation of the optical and mechanical properties of nanostructures (using e.g. Finite element methods).

We are seeking applicants to support development of exciting new methods to measure the condition of habitats and their natural processes (e.g., grazing, hydrology etc.). The currently known micropollutants in wastewater are only the ‘tip of the iceberg’ as they typically account for less than 5% of the total toxicity. Most solid materials are heterogeneous at the µm scale. The heterogeneity can stem from the subdivision into domains, e.g. crystals in polycrystalline metals, as well as from the presence of secondary phase particles and porosities. This second type of heterogeneity is critical, because it dramatically affects the material mechanical properties. The effect can be either beneficial and purposely pursued, like in particle-reinforced composites and porous orthopedic implants, or heavily detrimental and cause for unpredicted failures, like undesired porosity in 3D printed metal parts. Theoretical tools exist to predict these effects, but they are highly computationally demanding as they involve the numerical solution of nonlinear differential equations.

21. Postdoctoral Positions in characterization of CD109’s structure and interactions
Summary of Postdoctoral Fellowships:
CD109 is a GPI-anchored cell surface protein that modulates TGFβ signaling through direct interactions with TGFβ cytokines and the type I receptor TβRI. As a member of the α-macroglobulin protein family, which also includes complement factor C3 and human alpha-2-macroglobulin, CD109 has a complex multi-domain structure and can exist in multiple conformations. We have established the production of recombinant soluble CD109 in mammalian cells, as well as methods to induce its different conformations. This project will therefore be able to focus on the structural determination of CD109 by cryo-electron microscopy. The recombinant CD109 will also be used for binding studies with CD109’s interaction partners, as well as the identification of new interaction partners e.g. by pull-down studies.

Summary of Postdoctoral Fellowships:
Our group has invented a new technology that can convert existing antibodies into prodrug versions that must be proteolytically activated before they can bind to their target antigen. Until now, we have prioritized immune checkpoint blockade antibodies. This project, funded by the Danish Cancer Society, will investigate the application of our technology toward bispecific T cell-redirecting antibodies. Much of the proof-of-concept antibody engineering is already in place, and we anticipate that most of the research period will be used on antibody characterization and optimization. The candidate will participate in planning and preparing for in vivo experiments (in xenograft mouse models) testing anti-tumor efficacy, which will be performed by our collaborators at the CCIT. Summary of Postdoctoral Fellowships:
Cytoplasmic aggregates can be selectively eliminated by autophagy through a selective process called aggrephagy. This is particularly relevant in the nervous system where an impairment in autophagy leads to neurodegeneration and may explain why mutations in ATG genes cause neurobiological diseases. This clearance capacity of autophagy has unique therapeutic potentials because genetic and pharmacological activation of this pathway delays the onset of neurodegenerative disorders characterized by cytoplasmic aggregates such as Huntington’s, Alzheimer’s and Parkinson’s diseases. The mechanism underlying aggrephagy in non-neuronal and neuronal cells, however, remains largely unclear. In non-neuronal tissues, cytoplasmic aggregates are ubiquitinated and recognized by autophagy receptors such as TAX1BP1 and p62, which mediate their sequestration into autophagosomes. How this recognition is accomplished in neurons is unclear.

The work aims at clinical validation of non-invasive liquid biopsy tests for cancer biomarkers, analytical validation of the developed bioelectronic immune-tests and their further adaptation for lateral-flow immunoassays for POCT. Our aim is to complement radiological screenings and the histological and histochemical tests for cancers patients requiring life-long follow-up screenings, for whom a risk of not detecting all reoccurring tumors exists. The work is performed in a very close collaboration with the Aarhus University Hospital and is funded by the Novo Nordisk Foundation.
