The Grandma Karl research environment targets several aspects of pseudonymization, aiming to advance Sweden’s work on open access to research data. The primary focus of this position will be development of algorithms to automatically detect, label and pseudonymize personal identifiers in freely written texts (essays/blogs), focusing on linguistic challenges such as spelling errors, ambiguous entities, semantic constraints etc. A minor part of the research can be devoted to the analysis of type and number of personal identifiers versus acceptable protection, followed by reidentification tests to ensure that pseudonymization is effective.

3. Fully Funded PhD Position in Prevention of type-2-diabetes after gestational diabetes

Summary of PhD Program:

Gestational diabetes (GDM) may have serious short- and long-term consequences for both woman and child. Although glucose levels generally normalise after delivery, GDM increases the risk sevenfold for type-2-diabetes (T2D) within a ten-year period. Several studies show that the T2D risk after GDM can be reduced by lifestyle interventions and that early detection of impaired glucose tolerance is of great importance. There is currently, however, no satisfactory follow-up program after GDM leading to missed cases and delayed diagnosis of T2D. The objective of the PhD project is to measure and determine behavioural and biological factors important for disease progression from GDM to IGT/T2D, and to intervene by internet-based lifestyle modifications to reduce identified risks.

5. Fully Funded PhD Position in Molecular mechanisms of Alzheimer’s disease

Summary of PhD Program:

Alzheimer’s disease (AD) is one of the most common neurodegenerative diseases and it is estimated that more than 100,000 people are affected in Sweden. Research over the last 30 years has shown that the pathological changes in the brain can be attributed to different protein aggregates composed of amyloid-β (Aβ) and tau protein in the form of plaques and tangles, respectively. Studies have shown that in AD there is a very strong association between tau aggregates and Aβ-containing plaques. However, neither the mechanism behind tau aggregation nor the actual connection between Aβ and tau is currently known. It is not known how these processes lead to AD symptoms or what triggers the disease.

6. Fully Funded PhD Position in B cells in Neoehrlichia mikurensis infection

Summary of PhD Program:

Mononuclear cells from patients with malignant B-cell lymphomas, with and without N. mikurensis infection, are stimulated with bacterial and endothelial antigens and phenotyped regarding 23 different B-cell markers, and the generated multiparameter data is analyzed by Helios mass cytometry and cluster analyses. Comparisons are made with the B- and T-cells responses evoked by the infection in patients with normal immune defenses and without malignant B-cell lymphoma. In silico analyses of the N. mikurensis genome, bacterial extracts, and recombinant bacterial proteins and peptides are employed to develop high-throughput antibody diagnostics for N. mikurensis. The image flow cytometry method for antibody analysis is used for comparison.

Clonal hematopoiesis (CH) is an early stage of blood cancer development, where a large fraction of blood cells originates from a single hematopoietic stem cell lineage. CH can be detected in DNA sequencing data from blood cells of healthy individuals based on somatic genetic variants. CH carries an increased risk of leukemia and age-related diseases. Recently, we distinguished two groups of clonal hematopoiesis and stratified risk of developing distinct types of blood cancers (Niroula et al., Nature Medicine 2021). In this PhD project, we aim to identify new genetic variants underlying CH and identify regulators of CH development. The findings will be used for predicting the risk of developing blood cancer. This data-driven project will involve large-scale analysis of next-generation sequencing (NGS) data.
