Summary of Postdoctoral Fellowship:

The overarching goal of our research program is to better understand the molecular and cellular mechanisms underlying disease progression and drug resistance in hematological malignancies. Our group is interested in studying oncogenic signaling pathways and metabolic reprogramming in hematological malignancies. We aim to uncover previously unrecognized vulnerabilities for therapeutic intervention.

Our research involves multi-disciplinary approaches including molecular biology, biochemistry, chemical biology, immunology, mouse genetics and CRISPR-based gene editing. Recent relevant papers include Chan et al. Nature 2017 and Chan et al. Nature 2020. In the laboratory, we strive to promote a culture of inclusivity and provide mentorship on scientific growth and career development. The principal investigator will meet with the postdoc on a regular basis to review data, discuss goals and provide feedback and advice.

We are seeking an outstanding candidate with strong motivation to fill a post-doctoral position in the field of Molecular Cellular Immunology in Dr. Jayati Basu’s laboratory in the Department of Inflammation and Immunity. The postdoc will study molecular mechanisms regulating immune cell development and function in health and disease using mouse models and clinical samples. The research in the Basu lab focuses on development and functional lineage differentiation of lymphocytes and myeloid cell in thymus, bone marrow and barrier tissues.

Particularly, the lab studies how cytokine, chemokine, other tissue-derived factors and microbiota regulates transcriptional network to control development and function of these immune cells in heath and disease [Nat Immunol. 2021 Aug;22(8):969-982; Oncogene. 2023 Jan;42(5):351-363.; Commun Biol. 2022 Jan 21;5(1):84.; Nature. 2020 Jun;582(7810):109-114.]. Our lab is interested in molecular and cellular mechanisms responsible for smooth muscle cell (SMC) and endothelial cells (EC) phenotypic transitions. We use a combination of animal and cell culture models in combination with the bioinformatics analyses such as bulk and single cells RNA-sequencing to better understand these transitions, with the goal to develop novel therapeutic interventions for stabilizing atherosclerotic plaques in order to reduce the risk of plaque rupture with possible myocardial infarction or stroke.

The Xie lab at Lerner Research Institute at Cleveland Clinic is recruiting enthusiastic and motivated postdoctoral researchers to join us to study genome structure, dynamics and regulatory mechanisms in cancer and infection disease. We integrate cutting-edge super-resolution imaging (Xie et al., Nature Methods, 2020), live cell single molecule imaging (Xie et al., Genes & Development, 2017; Knight, Xie et al., Science, 2015) with modern genomics and genome engineering approaches to study the 4D architecture and dynamics of the mammalian genome (Xie et al., Mol.Sys.Bio, 2021) and the fundamental genome regulatory mechanisms in cancer evolution and infection diseases (King*, Yost*, Xie* et al., Nature, 2021).

We seek to understand how the epigenetic network (e.g., writers, reader, effectors), particularly the BET family proteins, regulate the basic genome structure and function (Xie et al, Nature Genetics, 2022, in press) in human development and diseases with the hope of discovering novel druggable targets to improve cancer therapeutics and to fight against infection. We are also interested in tool development to label, image and manipulate the human genome to improve disease diagnosis and in dissecting the interplay between genome regulation and other fundamental cellular processes (e.g., immune response).
