Two postdoctoral researcher positions are available in the laboratory of Dr. Xiaolu Yang at the Perelman School of Medicine at the University of Pennsylvania. The Yang lab studies cancer and neurodegenerative diseases. Our current projects focus on two broad areas:

(1) The tumor suppressor p53, metabolism, and autophagy. We are interested in the regulation and functions of the preeminent tumor suppressor p53. Our results have revealed a role for p53 in modulating metabolic pathways that are critical for biosynthesis and redox balance. We are investigating the function of p53 as both a sentinel and a regulator for metabolic activities. We are also identifying and characterizing metabolic alterations that drive tumor initiation and progression. A recent extension of this research area is to define the role of metabolism and autophagy in stem cells, including embryonic stem cells and cancer stem cells.

(2) Protein quality control, aging, and neurodegeneration. Our lab recently identified two protein quality control (PQC) systems, which consist of tripartite motif (TRIM) proteins and poly-Asp/Glu (polyD/E) proteins, respectively. Unlike canonical PQC systems, these new systems are independent of ATP and are unique for animals (the TRIM system) or eukaryotes (the polyD/E system). Both systems are multifunctional and highly effective. We are investigating their mechanisms of action; their roles in aging and neurodegenerative diseases, including Alzheimer’s disease, Parkinson’s disease, and amyotrophic lateral sclerosis; and their utility in treating these diseases.

The Heller Lab studies the mechanisms by which epigenetic remodeling regulates gene function, in the context of neuropsychiatric diseases such as addiction and depression. To approach this problem, we directly manipulate histone and DNA modifications at specific genes in vivo, using viral delivery of novel epigenetic editing tools, such as CRISPR/dCas9-fusion proteins. This ‘bottom-up’ approach allows direct elucidation of the causal relevance of epigenetic remodeling in the brain. The Romano Lab at the University of Pennsylvania is recruiting a postdoctoral researcher to develop and apply new methods in computational toxicology using artificial intelligence and techniques from biomedical informatics. We develop knowledge graphs to computationally predict and explain the downstream clinical effects that arise following exposure to environmental toxicants. This postdoc position will involve extending these knowledge graphs to new biomedical data types, and applying graph machine learning (Graph ML) to detect novel adverse outcome pathways. A position is available in the Mitchell Laboratory of Neurodegeneration for a postdoctoral student to investigate the relationship between lysosomal pathology and neurodegeneration. The research is translational and will focus on identifying pharmacologic and nanoparticle approaches to restore lysosomal function using cell physiology and molecular biology techniques. A PET imaging companion diagnostic will be developed in parallel as a prognostic biomarker as well as for image-guided treatment planning. Studies will utilize preclinical cell line and murine models to evaluate this highly translational theranostic approach. The Nathanson laboratory invites applications for Post-Doctoral positions at the Perelman School of Medicine, University of Pennsylvania. We focus on cutting-edge translational research in cancer genetics, both in the inherited and somatic genetics of cancer (example publications include Hu et al., NEJM 2021; Maxwell et al., JCO Pres Onc, 2020; Kraya et al., Clin Cancer Res 2019). This ‘bottom-up’ approach allows direct elucidation of the causal relevance of epigenetic remodeling in the brain.
